First obtain a list of all the files the spyware created and runs on your computer. You can do this by running spysweeper/spybot and looking at the files it wants to delete but can't. You could also try Hijack This, but I've never used it.
Boot in safe mode, or to a separate operating system, or a OS on cd-rom (slax or any rescue disc).
Delete all of the spyware files.
Reboot to normal mode.
Run spysweeper/spybot to clean the registry entries and any other leftover files that the spyware made.
That's how I usually do it when people ask me to remove the garbage they got on their computer.
Backup your system first if it makes you nervous.
