Actually I've tried using additional buttons on my mouse and I've yet to find anything useful for them to do that doesn't just obfuscate the overall interface.
I could imagine it being useful in MMO's where they usually don't care how much they fuck up the interface so they just shove more shit on the screen, though.

still puts more options at your fingertips instead of having to move your hand across a keyboard.
it's faster, especially when you can bind item slots and skills to the buttons
